The result: the light-induced change in the charge state of amino acid glutamate 90 (E90) triggers an increased penetration of water molecules, so that the protein can now purposefully conduct ions through the cell membrane.
Three methods combined
Using time-resolved infrared spectroscopy, the RUB biophysicists Jens Kuhne and Dr. Erik Freier have been able to show for the first time that the channel is opened through the deprotonation of the amino acid glutamate 90 (E90). In addition, the electrophysiological experiments of the researchers in Berlin confirm that a mutation of the amino acid leads to a change in the ion permeability of the protein. Instead of using safety goggles and lab coats, the two biophysicists Kirstin Eisenhauer and Dr. Steffen Wolf at the Department of Biophysics used supercomputers to simulate how the protonation change of the glutamate opens the channel and allows water molecules to penetrate.
Internationally preeminent

Gallup?s positive findings are echoed by the latest Citibank small business survey, which shows small business owners? hiring intentions at the highest they?ve been in two years. Over 25 percent of the small businesses surveyed intend to increase their permanent full time employees this year. That?s a 12 percent increase from January 2011 and a 9 percent increase in the last six months. In addition, 22 percent plan to bring on full time workers on a seasonal basis.
The majority (71 percent) of those surveyed in the Citibank report plan to keep their workforce the same size, but that doesn?t mean their outlook is negative. Forty-four percent say they?ll work longer hours themselves, and 40 percent expect their current employees to be more productive?all good signs that business is picking up, even if hiring plans haven?t yet kept pace.
But are small business hiring plans just pie in the sky? Small business owners in the Gallup poll were significantly more negative when asked to reflect on how much hiring they actually did in the past year. In 2011, about 22 percent decreased jobs while 13 percent increased them. The Gallup report says:
?Since the 2008 global economic collapse, the net percentage of small-business owners? self-reported actual job growth versus job cuts has generally remained negative, even when small-business owners say they are more likely to grow than contract.?
I don?t think these plans are pipe dreams, but I do think small businesses are waiting for the right time to commit to hiring. The results of the Citibank survey back me up: Overall, small business owners are confident, with 87 percent saying they think 2012 will be better than or the same as 2011. Nearly half say they are ?poised to grow when conditions are right.? And another 33 percent believe their businesses will be growing rapidly or moderately in 2012.

Clementi's story touched off a national discussion about bullying of young gays after he committed suicide in September 2010 by jumping off the George Washington Bridge, days after the intimate encounter.
Judge Glenn Berman said Tuesday that most of the roughly 190 prospective jurors who filled out questionnaires last week said they had heard about the case.
He dismissed those Middlesex County residents who indicated that they had made up their minds.
Roommate of gay Rutgers student reportedly sent him conciliatory texts
Also on Tuesday, Berman fixed an error that could have had an impact on the trial: A hindering apprehension charge was changed from a third-degree crime to a second-degree crime.
Normally, that technical change would mean that a defendant who's found guilty would likely face a prison sentence. But Berman said he could still view that charge as a third-degree crime for sentencing purposes if Ravi is found guilty of it. He said he would decide that issue later if he needs to.
Charged with invasion of privacy, not with death
Ravi has not been charged with causing Clementi's death. Instead the main charge is invading Clementi's privacy, which is unlikely to result in prison time for a first offender. For a hate crime conviction, which carries a sentence of up to five to 10 years, prosecutors must prove Ravi attempted to intimidate Clementi for being gay. Ravi also faces related charges of tampering with witnesses and evidence.
Ravi has pleaded not guilty to all the charges, and has rejected a plea deal with prosecutors that could have helped him avoid a prison sentence. Instead he now risks up to 10 years in prison if convicted.
"Most hate crimes are committed against strangers, not against roommates or against good friends or neighbors," Jack Levin, a professor of criminology at Northeastern University, said in an interview.
Successful prosecutions typically rely on racial or homophobic slurs made as the crime is committed, graffiti left at the scene, the possession of hateful propaganda by the offender, or an offender's previous history of hate crimes.
"In this case there apparently are none of those indicators to be found," Levin said. "It doesn't sound like a very strong case."
On Sept. 19, 2010, Clementi had arranged to have a man in his mid-20s identified in court only as M.B. to come over, and asked Ravi if he would leave the room. Ravi agreed. He went to the room of Molly Wei, a friend across the corridor, and used her computer to access his webcam through video-chatting software.
Wei later told investigators Ravi was alarmed at having to leave his room for a visit from an unknown older man and wanted to know what was going on. She said they saw images of Clementi kissing M.B., and, shocked, turned the video feed off within seconds. Ravi posted a message on his Twitter account:
"Turned on iChat and saw my roommate making out with a dude. Yay."
Defense points to Clementi's jokes
To secure the bias intimidation conviction, prosecutors will need to convince a jury Ravi invaded Clementi's privacy and that he did so to intimidate him because he was gay.
Ravi's lawyers have pointed to transcripts of Ravi's online conversations with friends showing that, after the gossip value of discovering his roommate's sexuality has apparently faded, he concludes that Clementi being gay is "no big deal."
Ravi's lawyers have said in court documents there is no evidence Clementi was intimidated by Ravi's actions. He was open about his sexuality with friends and had come out to his parents, according to Clementi's chat transcripts.
After learning from Ravi's Twitter post he had been seen on the webcam, Clementi said in an online chat with a friend he felt "violated" at first, but soon after joked about the incident, saying he found it "sooo funny," and laughed when the friend suggested it could be considered a hate crime.
Nonetheless, he filed a formal request to switch roommates and posted his concerns on an Internet forum for gay men, which prosecutors are likely to point to as evidence he felt intimidated.
Even after realizing his encounter with M.B. had been briefly viewed by Ravi, Clementi invited M.B. back to his room two days later and again asked Ravi to excuse himself, although this time he was careful to make sure Ravi's computer was unplugged.

Hi all, Have a problem where the end bay of my garage floods. I have a drain along the front though it is not big enough to handle heavy rain. Looking at concreting a hump on the outside to prevent the water, only needs to be a couple inches high. See photo , I have drilled in some reo bars and tied some across to help with strength. Any experience out there to help me . Thanks |

Can you post a picture looking from the garage out across the drive, and same from the top of the drive down to the garage, so you show the slope of it. I would have to say from all appearances, that drain width is either clogged with stone, or too small, if it is not taking on the water from an extreme downpour. A concrete hump is not going to do anything, since the rain when it falls, will still go into the garage. Hi thanks for comments. Agree in hindsight the drain is too close... You can see from this photo that it is a steep driveway and if I'd have ramped away from the shed it would've made it even steeper (but maybe a good option compared to water). Mixed with the end bay being 65mm lower than than the rest of the shed (this was originally an open bay and the shed company said that would weatherproof the enclosed bays. Cheers |

Yep, you are funneling all of the water coming down the hill into the shed. I would say, that you need to redo the drive, so that the water goes elsewhere, vs. being directed down to the bottom to pool. You could always sink a couple of storm sewer drains at the bottom, placing it just high enough that the rock does not wash into it, but it would allow storm water to drain. __________________Ren: Now listen, Cadet.
